dubbelresistent PDX-modell (patient-derived xenograft-modell) med celler Establishment of an in vivo mouse model to study and overcome
Establishing a robust and reliable animal model is the foundation for studying the pathogenesis of cancer. Establishing xenograft tumor models from patient-derived tumor tissue (PDTT) at low passage is believed to conserve original tumor characteristics such as heterogeneous histology, clinical biomolecular signature, malignant phenotypes and genotypes, tumor architecture, and tumor vasculature. Patient-derived xenograft models (PDXs) PDXs are aiming at addressing the shortcomings of human cell line derived xenografts and are increasingly complementing or even replacing them. Breast cancer is a heterogeneous disease and tumor heterogeneity across patients as well as intratumoral heterogeneity hampers treatment decisions and success. Xenograft, Orthotopic & Syngeneic Models WBI offers xenograft, orthotopic and syngeneic models for assessment of anti-cancer agents. The following are selected cancer models: Breast Cancer Models (BT-474 & MDA-MB-231) Abstract Patient-derived tumor xenografts (PDXs), in which tumor fragments surgically dissected from cancer patients are directly transplanted into immunodeficient mice, have emerged as a useful model for translational research aimed at facilitating precision medicine. Patient-Derived Human Xenografts PDTX models or “xeno-patients” are made by implanting cancerous tissue from a human primary tumor into an immunodeficient mouse.

CrownBio offers a comprehensive program for xenograft and syngeneic model establishment. PANC-1 Cell Line Derived Xenograft. An assortment of human xenografts, such as the PANC-1 CDX model (cell line derived xenograft), are established by injection of cancer cells (subcutaneous, intraperitoneal or intravenous) into immunocompromised mice. KARPAS 299 Cell Line Derived Xenograft. In vivo xenograft tumor models are instrumental in addressing mechanism of action (MoA), PK/PD and evaluating combination therapies. KARPAS 299 cells were initially isolated from a patient with T cell lymphoma and are grown under asceptic conditions prior to subcutaneous injection in immunocompromised mice.
New medicines are tested on staged tumor growths that have been engrafted via subcutaneous or orthotopic inoculation in an immunocompromised mouse or rat model.
These models are reported to reflect the underlying biology and heterogeneity of individual tumors more accurately than conventional cell-line-derived xenograft (CDX) models . In addition, drug response of PDX models often replicates the clinical response to the same drug in individual donor patients [ 7, 8 ].

In oncology, these have traditionally been cell line xenograft (CDX) models using tumor cells immortalized in vitro from patient tissues. Xenograft: A surgical graft of tissue from one species to an unlike species (or genus or family). A graft from a baboon to a human is a xenograft.

A cell line-derived Xenograft or CDX model is used for the research and testing of anti-cancer therapies. Human tumor samples are cultured as cell lines and implanted into immunodeficient nudes to test the efficacy of anti-tumor compounds in vivo. This VM xenograft model provides a reliable platform to investigate the cellular and molecular mechanisms driving VM formation and expansion. In addition, this model will be instrumental for translational studies testing the efficacy of novel drug candidates in preventing the abnormal vessel enlargement seen in human VM.

Lung cancer is the most common type of cancer worldwide, accounting for 13% of the total number of new diagnoses and is classified according to histological type by the size and appearance of cells: non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC).

These models can then be maintained by successive passaging through generations of xenograft mice, resulting in an easily generated and consistent platform for preclinical and mechanistic studies. To effectively evaluate novel anticancer agents to treat RMC, an established PDX model is first needed. 2020-03-01

With Syngeneic models, the tumor cell line is derived from the same species as the host animal. Syngeneic models can thus test the efficacy of immuno-oncology agents as single therapies, or in combination with other anti-cancer drugs. These models are reported to reflect the underlying biology and heterogeneity of individual tumors more accurately than conventional cell-line-derived xenograft (CDX) models .

To study the underlying mechanisms, we induced therapy resistance in patient-derived xenograft (PDX) models of BRCA1-mutated and BRCA1-methylated triple-negative breast cancer. Methods: A cohort of 75 mice carrying BRCA1-deficient breast PDX tumors was treated with cisplatin, melphalan, nimustine, or olaparib, and treatment sensitivity was determined.
